hypertension of chronic hypoxia (part 4)

Chronic hypoxia: Four groups of rats were used, and adrenalectomy was performed in two of them. Three days after surgery, one adrenalectomized and one sham-operated group were exposed to isobaric hypoxia (fraction of inspired oxygen of 0.1) for two weeks. The other two corresponding groups were kept in room air. Chronic hypoxia was induced in the isobaric hypoxic chamber by a slight modification of a previously described system . The chamber was opened three times a week to clean the cages and supply food and water or saline solution for drinking. After two weeks of hypoxia the rats were removed from the chamber and anesthetized with pentobarbital (25 mg/kg of body weight, intraperitoneally), and the pulmonary artery was cannulated with a specially shaped polyethylene catheter.Pulmonary arterial blood pressure was measured in rats spontaneously breathing room air. In addition, in the adrenalectomized and sham-operated rats not exposed to hypoxia, pulmonary artery blood pressure was measured during a 20 min period of breathing 10% O2 + 90% N2 gas mixture. Then the rats were killed by an additional dose of pentobarbital, the chest was opened, the heart was dissected, and the right and left heart ventricles and heart septum were separated and weighed. Lungs were fixed with neutral formol solutionby infusion at a hydrostatic pressure of 12 cm H2O through the tracheal cannula. Lung sections were stained by the hematoxylin resorcin fuchsin method, and the number of muscularized distal pulmonary arteries was counted and expressed as a percentage of all respective vessels seen in the slide.
