Adrenalectomy in rats depresses hypoxic pulmonary vasoconstriction in vitro but does not attenuate the pulmonary hypertension of chronic hypoxia (part 2)

The control lung donor rats (but not the control blood donors) were subjected to sham adrenalectomy. All groups of rats were fed standard diet; adrenalectomized rats were given 1.0% NaCl solution instead of drinking water. After adrenalectomy the animals were monitored for visible signs of inflammation or other complications, but none were encountered. There was no mortality after surgery or during exposure to hypoxia.  Isolated rat lung preparation: Acute effects of hypoxia and angiotensin II were studied in the isolated, perfused rat lung preparation . Five to 10 days after adrenalectomy or sham operation the lung donors were anesthetized with pentobarbital (30 to 40 mg/kg of body weight, intraperitoneally).After a thoracotomy and intracardiac injection of heparin (100 U) the pulmonary artery, left ventricle and trachea were cannulated. Then the lungs were suspended in a humid and warmed chamber. They were ventilated with a humid gas mixture of 21% O2 + 5% CO2 + 74% N2 (normoxic gas) at 60 breaths/min. The inspiratory pressure was 9 cm H2O and end-expiratory pressure was 3 cm H2O. The lungs were perfused through the pulmonary arterial cannula by a peristaltic pump with blood obtained from blood donors.
